Glycogenin is a sterol and is found in the plasma membrane of a variety of cells. While key components of the endomembrane system, glycogenin is a versatile, circulating protein which, together with glycogen, is a fundamental energy buffer. In addition to the glycogenin present within the plasma membrane, glycogenin is also present in the cytoplasm and in many places in the nucleus, cytoskeleton, Golgi and mitochondria. Human glycogenin is encoded by NAGLU, a single copy gene of approximately 1.4kb and is translated with a 40 amino acid mitochondrial leader sequence. Glycogenin is present in the liver and muscles and at very low levels in the brain. Muscle and liver analyses indicate glycogenin is decreased in DM1 patients [9]. Muscle biopsies have demonstrated expression of glycogenin in the plasma membrane, in addition to the expected expression in the nucleus, cytoskeleton and mitochondria [9]. These findings suggest that the common liver and muscle glycogenin gene mutation reduces expression levels of the protein (greater than twofold) [9]. The normal function of glycogenin within the muscle cell is not definitively understood, however glycogenin has been implicated in insulin signaling, regulation and adhesion. In particular, glycogenin may play a role in cardiac differentiation, as it is expressed during cardiac development and also in response to insulin. While the function of glycogenin in the heart is not clear, recent animal studies have indicated glycogenin function in embryonic development. In particular, mice lacking the glycogenin gene have demonstrated increased growth rate, decreased adipose tissue and increased bone mineral density and muscle mass. Glycogenin-deficient mice also have no circulating antibodies against insulin and in contrast to wild-type mice, are also deficient in circulating antibodies against insulin. Interestingly, they develop hyperinsulinemia, hyperglycemia, progressive type 2 diabetes mellitus, increased body weight and reduced adiposity. In addition, glycogenin null mice have been recently shown to have increased skeletal muscle mitochondrial density and function, increased sensitivity to insulin and increased insulin-stimulated glucose uptake [9]. This work indicates that glycogenin’s function is an important in insulin sensitivity and that absence of the protein can have major consequences, including increased insulin sensitivity and growth rate and other metabolic abnormalities that are often seen in DM1 patients. d2c66b5586
